Horseradish peroxidase (HRP) is an indicator enzyme often used for labeling to to antibodies and other non-colorogenic proteins as a color indicator. It is used in ELISA, Western blotting and Southern blotting assays.  For labeling antibodies in ELISA. RZ (= A 403/A 275) = 3.0

Unit definition: 1 U catalyzes the decomposition of 1 µmole hydrogen peroxide per minute at 25 °C, pH 7.0; reaction coupled with phenol- aminoantipyrine.


Activity in other units: min. 250 purpurogallin units/mg (1 purpurogallin unit catalyzes the oxidation of 1 mg pyrogallol to purpurogallin in 20 seconds at 20 °C and pH 6.0.


The purpurogallin is extracted and determined spectrophotometrically at 420 nm.

1 mg purpurogallin requires 13.5µmole peroxide, 1 purpurogallin unit corresponds to the decomposition of 12 µmoles peroxide at 25 °C).
